چکیده

frequencies of the genetic polymorphisms in CYP3A4 (10% heterozygous for CYP3A4*1B, 5.4% heterozygous for CYP3A4*2, and 2.2% heterozygous for CYP3A4*3), this implies that ϳ15% of the (Caucasian) population may carry a genetic polymorphism in this allele. Because genetic polymorphisms may exhibit strong differences in occurrence among different ethnic groups, other populations need to be investigated to determine the allelic frequency of CYP3A4*3. In conclusion, we have described and validated a PCR-RFLP assay for the CYP3A4*3 allele. The frequency of this variant allele in the Caucasian population (1.1%) indicates that it might be important in predicting CYP3A4 activity based on genotype. Future research should be directed toward elucidating the effect of this polymor-phism on CYP3A4 enzymatic activity and toward establishing whether this is solely a genetic, or also a functional , polymorphism. NO is produced in many different cells and is involved in the regulation of such physiological and pathophysiolog-ical processes as inflammation, vasodilation, and metabolism (1). Depending on the cell type, NO is produced in an enzymatic reaction catalyzed by one of the three isoforms of NO synthase (NOS): neuronal NOS, endothe-lial NOS, and inducible NOS (2). Measurement of the NO metabolites, nitrate and nitrite (NOx), is also important as a marker of NOS enzyme activity. Obesity is a condition involving an excess accumulation of body fat, and the prevalence of obesity is rapidly increasing worldwide. Excessive weight and obesity are leading …

Overweight and Obese Humans Demonstrate Increased Vascular Endothelial NAD(P)H Oxidase-p47 Expression and Evidence of Endothelial Oxidative Stress

Background—Obesity may alter vascular endothelial cell protein expression (VECPE) of molecules that influence susceptibility to atherosclerosis. Methods and Results—Quantitative immunofluorescence was performed on vascular endothelial cells collected from 108 men and women free of clinical disease who varied widely in adiposity (body mass index 18.4 to 36.7 kg/m; total body fat 5.8 to 55.0 kg; ...

Clinical implications of thermal therapy in lifestyle-related diseases.

Systemic thermal therapy, such as taking a warm-water bath and sauna, induces systemic vasodilation. It was found that repeated sauna therapy (60 degrees C for 15 min) improved hemodynamic parameters, clinical symptoms, cardiac function, and vascular endothelial function in patients with congestive heart failure. Vascular endothelial function is impaired in subjects with lifestyle-related disea...

طراحی تاثیر تمرینات هوازی بر سطح پلاسمایی اکسید نیتریک و عملکرد اندوتلیوم عروق زنان یائسه

Background: Cardiovascular disease is one of the main causes of morbidity and mortality in postmenopausal women. Physiological decrease in estrogen levels and accumulation of visceral fat and being overweight, increase the risk of cardiovascular disorders in the menopausal years. The aim of this study was to determine the effects of aerobic exercising on plasma nitric oxide level and ...

Effects of obesity on endothelium-dependent reactivity during acute nitric oxide synthase inhibition: modulatory role of endothelin.

This study investigated vascular reactivity in response to acetylcholine, in the presence of acute inhibition of nitric oxide synthase, in the carotid artery and aorta of obese C57Bl6/J mice fed on a high-fat diet for 30 weeks, and of control mice. A subgroup of obese animals was also treated with the ET(A) receptor antagonist darusentan (50 mg x kg(-1) x day(-1)). In vascular rings from contro...

Regulatory role for the arginine-nitric oxide pathway in metabolism of energy substrates.

Nitric oxide (NO) is synthesized from L-arginine by NO synthase in virtually all cell types. Emerging evidence shows that NO regulates the metabolism of glucose, fatty acids and amino acids in mammals. As an oxidant, pathological levels of NO inhibit nearly all enzyme-catalyzed reactions through protein oxidation. However, as a signaling molecule, physiological levels of NO stimulate glucose up...
